What Is FuGENE® HD Transfection Reagent?
FuGENE® HD is a non-liposomal, multi-component transfection reagent designed to deliver nucleic acids into eukaryotic cells with high efficiency and low toxicity. It is manufactured without animal-derived components and is compatible with serum-containing and chemically defined media. This makes it suitable for applications where physiological relevance and reproducibility matter — from basic research to protein and virus production.
Transfection reagents vary widely in how much they stress the cells they deliver into. In a head-to-head comparison of four reagents, FuGENE® HD produced the fewest off-target transcriptional changes (Jacobsen et al., 2009).
Which Cell Types Can Be Transfected with FuGENE® HD?
FuGENE® HD supports transfection of a broad range of cell types, including many lines considered difficult to transfect. Validated cell types include:
- primary cells
- cancer model cell lines
- stem cells
- insect cells
The Transfection Assistant helps you optimize transfection protocols using information from published literature. Find citations on transfection of specific cell types, including the amount of nucleic acid and reagent-to-DNA ratio used.
Why Choose FuGENE® HD Over Other Transfection Reagents?
FuGENE® HD outperforms common lipid-based transfection reagents on two metrics that matter most for transfection: high efficiency and low toxicity.
Two practical advantages set it apart:
- No need to reduce serum before transfection. Cells can be transfected in up to 100% serum, so serum-dependent and primary cells can be transfected without first adapting them to low-serum conditions.
- Reagent/DNA complexes do not need to be washed away post-transfection. This reduces handling steps and stress on cells.
High Expression, Low Toxicity
| FuGENE® HD Benefits | Why it matters |
|---|---|
| Efficient DNA delivery | More cells take up your DNA, giving a strong signal for downstream assays |
| Low toxicity | Keeps cells healthy, so your results reflect your gene, not the reagent |
| Transfects in up to 100% serum | Works with serum-dependent and primary cells without switching them to low-serum conditions first |
| No wash or medium change after transfection | Fewer hands-on steps and less disruption to your cells |
| No animal-derived components; compatible with chemically defined media | Fits animal-free and chemically defined media workflows |
Unrivaled Transfection Performance
Transfection of Stem Cells
“FuGENE® HD resulted in the highest stable transfection rate, estimated by antibiotic selection, with plasmids containing genes under the control of the EF1alpha or PGK promoters”.

Frequently Asked Questions
Does FuGENE® HD require serum-free conditions for transfection?
No. FuGENE® HD works in serum-containing media and can transfect cells in up to 100% serum. There is no need to switch to serum-free media before or during the transfection step, which simplifies the protocol and maintains physiologically relevant culture conditions.
Is FuGENE® HD suitable for generating stable cell lines?
Yes. FuGENE® HD has been used to generate stable transgenic lines in multiple cell types, including human embryonic stem cells. In one study using a human ES cell line, FuGENE® HD gave the highest stable transfection rate among the chemical reagents tested when paired with EF1α or PGK promoter-driven constructs (Liu et al., 2009).
How do I find the optimal transfection conditions for my cell line?
The Transfection Assistant helps you optimize transfection protocols using information from published literature. Search by cell line name to find the amount of nucleic acid and reagent-to-DNA ratio used for specific plate formats.
