Quantify IL-12 Release Without Tedious Wash Steps

IL-12, a heterodimeric pro-inflammatory cytokine, is a key player in host defense through its ability to stimulate both innate and adaptive immune effector cells. IL-12 has emerged as a leading target for immunotherapy due to its role in innate and adaptive immune responses as well as antitumor effects.

The Lumit® IL-12 Immunoassay quantitatively measures released IL-12 in cell culture samples using a simple, no-wash protocol. Just add labeled antibodies to the sample, add detection reagent and read luminescent signal using a standard plate-reading luminometer. The entire protocol is completed in less than 70 minutes! The assay can be used directly on cells in culture or culture medium transferred to a separate assay plate.

How the Lumit® IL-12 Immunoassay Works

Primary antibodies to IL-12 were selected for their specific and sensitive detection and labeled with the LgBiT and SmBiT subunits of NanoBiT® Luciferase. In the presence of IL-12, the subunits are brought together to form an active luciferase enzyme. Addition of optimized substrate generates a bright luminescent signal proportional to IL-12 levels. The assay is specific for the IL-12p70 (A/B heterodimer); it is effective for detection of IL-12p70 in presence of 100-fold excess of either IL-12B subunit, IL-12p80 or IL-23.
