A Simple, Bioluminescent Assay for Glycosyltransferase Activity

The UDP-Glo™ Glycosyltransferase Assay provides a homogeneous, single-reagent-addition method to rapidly detect UDP formation in glycosyltransferase reactions. After the glycosyltransferase reaction, an equal volume of UDP Detection Reagent is added to convert the UDP product to ATP and generate light in a luciferase reaction. The light generated is correlated to UDP concentration by using an UDP standard curve.

This assay is intended for use with purified glycosyltransferases that use UDP-sugar as a donor substrate and cannot be used with whole cells or cell extracts. However, glycosyltransferases purified from cell extracts using immunoprecipitation or affinity tag pull-down can be used in the UDP-Glo™ Assay.

Applications of the UDP-Glo™ Assay include profiling glycosyltransferase specificity for different sugars, screening ]library compounds for effects on glycosyltransferase activity, and detecting chemical compound glucuronidation during drug discovery.

UDP-Glo™ Glycosyltransferase Assay Reaction: Following the glycosyltransferase reaction, free UDP is converted into ATP, which is then converted into a luminescent signal that is proportional to the glycosyltransferase activity in the reaction.

Works with Most Glycosyltransferases

UDP-Glo™ Assay can be used to measure the activity of just about any glycosyltransferase (GT) that uses a UDP-sugar as a substrate. Here, we looked at GTs involved in glycan biosynthesis, post-translational modification and drug metabolism.

Detects Low Nanomolar Amounts of UDP; Ideal for Low-Activity Glycosyltransferases

The assay response is linear in the nanomolar to micromolar range, allowing use of low concentrations of UDP-sugar, and alleviating issues due to feedback glycosyltransferase inhibition.

The data to the left shows the linearity and sensitivity of the UDP-Glo™ Glycosyltransferase Assay. Here, a UDP standard curve was prepared over the indicated range of UDP concentrations in 25µl of 1X glycosyltransferase reaction buffer in a solid white 96-well plate. UDP-Glo™ Glycosyltransferase Assay was performed using 25µl of UDP Detection Reagent at room temperature. Luminescence was recorded using a GloMax® luminometer. Values represent the mean of four replicates. The table shows luminescence measured 1 hour after adding the UDP Detection Reagent, and signal-to-background ratios calculated for each concentration of the UDP standard curve.
Standard curve using UDP concentrations from 0.02 micromolar to 25 micromolar.

Luminescent Signal is Stable Over Several Hours

The UDP-Glo™ Assay  generates a highly stable luminescent signal with >80% signal remaining after 3 hours, allowing convenient batch-processing of assay plates.

The data here shows that the signal obtained with the UDP-Glo™ Assay is stable for several hours. To determine signal stability, luminescence was recorded every hour. The signal-to-background ratio did not change over the time measured (data not shown). RLU = relative light units.
