Express Regulatory Proteins in Mammalian Cells

The CCNT1 Expression Vector contains the coding region of the human CCNT1 regulatory protein. The expression vectors are designed for use with kinase-NanoLuc® Fusion Vectors that contain the coding region of a kinase-NanoLuc® (or NanoLuc®-kinase) fusion protein. The Expression Vector and kinase-NanoLuc® Fusion Vector are cotransfected into cells prior to analysis using the appropriate NanoBRET® Target Engagement Intracellular Kinase Assay. These regulatory protein Expression Vectors are supplied transfection-ready, with a CMV promoter that drives expression of the full-length regulatory protein.

NanoBRET® Target Engagement (TE) Intracellular Kinase Assays

The NanoBRET® TE Intracellular Kinase Assays can analyze the affinity of test compounds by competitive displacement of a fluorescent NanoBRET® Tracer reversibly bound to a NanoLuc®-kinase fusion protein in cells. Introduction of competing test compounds results in a dose-dependent decrease in the NanoBRET® signal for estimating intracellular affinity against the target protein. The NanoBRET® Tracer used is determined by your choice of NanoLuc® kinase fusion vector. The Kinase Vector Selection table shows which NanoBRET® Tracer assay is correct for each kinase fusion vector.
